The number of proliferating cells in the dorsal thalamus was examined in mouse brains, with particular emphasis on the pattern of foci that bind to proliferating cell nuclear antigen (PCNA) in the diencephalon. To study the growth patterns of the dorsal thalamus, bromodeoxyuridine (BrdU) was injected into mice at different postnatal ages. BrdU-positive cells were identified in the dorsal thalamus, but not in the ventral thalamus. The densities of BrdU-positive cells in the dorsal thalamus decreased from postnatal days 7 to 12 to day 15. The densities of PCNA-foci in the dorsal thalamus were significantly greater at day 8 compared to day 15. The number of BrdU-positive and PCNA-foci-positive cells at day 15 was greater than that at day 8. To examine the temporal relationship between BrdU and PCNA-foci, the brains of mice injected with BrdU at postnatal day 4 were examined on day 8 and day 15. PCNA-foci were detected in the preoptic area, but not in the posterior thalamus, from day 4 to day 15, whereas BrdU-positive cells were found in both the preoptic and posterior thalamus from day 8 to day 15. We also used antibodies against BrdU and phosphorylated histone H3 (pH3) to confirm the results of the PCNA foci. pH3 is a marker of mitotic cells and it was localized in the nuclei of the pyramidal cells in the preoptic area from day 7 to 15. On the basis of these findings, we propose that mitotic cells are mainly responsible for the increase in PCNA foci density from day 8 to day 15 in the dorsal thalamus, and that the preoptic area contributes to the mitotic cells that induce the expression of PCNA.Q:
